The insert contains the following sites separated by (kb)(approx): EcoRI- 1.5- BamHI- 1.2- HindIII 1.2- BglII- 0.5- EcoRI. Restriction digests of the clone give the following sizes (kb): EcoRI--4.4, 4.4; HindIII--6.3, 2.5; PstI--6.5, 2.5; BamHI--6.8, 2.0; BamHI/HindIII--6.0, 1.6, 1.1, 0.1; EcoRI/HindIII--4.4, 2.7, 1.7. Codominant segregation of the BglII RFLP was shown in 19 cystic fibrosis families (94 individuals) and 23 CEPH families (378 individuals). The insert is the 3' flanking region of the human carboxypeptidase A (CPA) gene. The 1.7 kb EcoRI/HindIII fragment is single copy and was used to detect the BglII RFLP.
